Building off existing concrete floors

I have a double garage and would like to convert one into another room. The dividing wall between the two garages is a cavity 4/3/4 block wall. There is a large opening in the wall approx 2 mts. I would like to cloe this gap, leaving just a normal door frame. There is a lintel over the gap so it would be just infill. The floor is 100mm thick.
The question is can I build off the floor with lightweight blocks, celcon or similar to fill this gap. I really don't fancy digging it all up.....!
