Many years ago I laid a patio in my back garden over a sand base.
I'm fighting a yearly battle with the dreaded weed !

Could I lift the slabs and lay a membrane over the sand and then re lay the slabs ?
Will this work ?

It will be as much work as when you original laid the patio.
Weedkiller once a year, be a lot easier
 
the weeds don't come from underneath, they grow in the jointing material which in your case is sand. Applying 'season long' Pathclear type products will mean it needs sprayed probably twice a year. The cheaper and better option is wash over the lot a few times a year with 1 part bleach 3 parts hot water and brush it on with a stiff brush.

Of course the bleach will kill most plants and grass so only suitable for area.s that are not edged in lawn or beds.
