When does an extension become a separate dwelling

If a 2 storey extension is built and a separate front door is built as long as there is a internal door leading.to the extention inside is that acceptable for.planning purposes

By "separate dwelling" do you mean "annexe"?
Are you trying to determine whether the extension falls under permitted development or needs planning permission?
