    Looks like condensation/hygroscopic grease stains. The stain will leech through paper. You need to stain block it.

    Seems you have some contamination withing the masonry. As said use a stain blocker before painting.

    You can't normally prop a purlin perpendicular to the rafters with vertical posts. The posts are not transferring the load.
