Yes it can be done, BUT it is not recommended if you can't find the stopcock.
Things can go wrong when fitting and will flood the place.
Best to find an isolation first and test to see if it works.
Really should be looking to fit a new stopcock first.
You need to remove a litre of water from the system first before you add a litre of X400. (drain a little more to give youself some extra space).
That is of course assuming that your only putting one, 1 litre bottle in.
No need to shut system down if the radiator valves are holding...

As has been mentioned before, by dormermike, (and others) add insulation where you can and make sure you have no draughts from places that there shouldn’t be any.
Round the windows are a favourite place for draughts, badly fitted...
You will have to empty a lot more water than just the water contained within the expansion tank (and also within the expansion pipe).
The chemicals probably will get through the complete system but will take a while to leach in the the main circulation.

Use the time to try and source a valve cheap. ebay for example. Your in no rush so don't need to bid high.

Any valve should last longer than that.
Is there corrosion inhibitor added?

Posible problem :-
The downstairs Myson valve sounds as if it is passing water through, this could be down to not moving its full travel to "off" position, or when it is fully off the water still gets through, thus heating your radiators.
If the valve is not in its fully closed position there...

Another posibility during a re-roof would be to nail an underfelt to the roof, then get a heavy grade felt, not the cheap "shed felt" though, use a cold applied bitumen...
