no the slave door opens second, the slave door will have a part screwed into it called a t-bar or lap bar, this is what the locking door locks into. the slave door needs to be fastened in by the shoot bolts fairly tightly.

does the lock work ok when the door is open? if so there is an alignment problem. sometimes it only takes a fraction to stop the lock engaging properly and stopping the key to turn fully.

if you take the handles off you should be able to see the lock mechinism, depending on type you may be able to see some sliders which operate when you move the handle. you might be able to move these with a screwdriver to see if it will move the mechanism. if not only other thing i can think of...

is it a standard head unit? if not most of the aftermarket ones are not compatible with the steering wheel controls. if it is the standard one then maybe the plug on the back has come loose.

bit of a strange one that, try taking the barrel out again and see if that turns freely with the key in if it does then its deffo a problem with the locking mechanism or you havent quite aligned the barrel up properly. i had a similar thing with a patio door locking mechanism, very difficult...
