The little black plastic handle I turn to release the water into the system seems very loose. I have looked and there is a screw in it, I have tried tightening the screw and loosening it but nothing is happening at all. Should I remove the screw?

I have a basi Garda M90F boiler, all seems to be working fine but to day when looking at it, the pressure seems to have dropped slightly. So I pulled out the manual, turns out my my filling loop is already connected so I opened the valve to fill slightly but nothing happened?? I have done...
