Oversite to suspended timber floor

I am going with a suspended timber floor to the extension. Once the blockwork is up what materials are acceptable to lay for the oversite. Am i correct in thinking its either 100mm or 50mm with a membrane underneath?

If there was access to back a wagon in i would have gone down the readymix route but I don't have that luxury. I would like to avoid mixing 2-4m3 of concrete and barrowing it round and in.

Am i able to use pea shingle/gravel/type1. If so is the 50mm depth correct?
