There is a special GAS version of ptfe tape but i wouldn't use it. What kind of a connection is it? i might use it on a compression fitting as a last resort. Use gas paste. Do you have a gas meter? Or is it bottled gas?

Anywhere between 1 and 1.5 bar is ok. Some boilers won't work at all if the pressure is below 0.7 bar (britony combi 80 etc)

get a CORGI to look at it. i'd do a test at the meter with your pilot light off. if the manometer is showing a drop, you have a leak somewhere on the system. if possible isolate all other appliances whilst doing this test.

As DP mentioned, its a 3 way valve/filling loop on this boiler and its the black plastic handle on the right hand side underneath your boiler. it should be pointing upwards at present, when its fully pointing downwards you will hear the water from the mains filling the boiler up. Watch the gauge...

The main water stopcock for the house will be at ground level unless you have a cellar, then it could be down there. God knows what you have found under the bedroom floorboards but it won't be the mains stopcock (unless maybe you live in a bungalow)?
