We have changed our bathroom to become a wet-room for our elderly dad.
The chair is fitted to the wall and folded when not in use.
There are 2 chair legs.
The chair leg unfolds directly onto the plastic drain cover.

We are concerned that the weight of a person will eventually cause the drain cover to break and is also a safety issue. The builders are refusing to amend the issue (

Any advice pls?

Given that is on the edge rather than the middle of the cover I don't think you have anything to worry about. That seat looks like the one we had in our shower, a plastic cheapo that quickly became covered in black mold that was difficult to keep clean in all the nooks and cranies. I think you will upgrade to a better quality one rather soon, at which time you can reposition it if you still have concerns.

The drain cover is raised above the floor level (and not touching the floor) - so the entire weight of the chair leg is on the drain cover

IMO it is down to the installing constructors to provide a finished job that is satisfactory and fit for purpose. An ill-fitting shower seat is not fit for purpose and should be addressed by the installer. The seat fixings either need moving over or a new wider seat provided before payment is made.
If in doubt, consult your area Trading Standards Office

Poor workmanship, I'd insist it is rectified. Should the waste break and/or give way, that could affect the stability of the chair and the person sitting in it. Can Chair be moved?
