Megaflo woes

Tonight our megaflo boiler started making a noise. We've listened all around the system as far as we can to try and pin the noise down but haven't been able to. The noise disappears when we switch the hot water to the off position, we normally have it set to constant.

My wife is panicking and wants to leave the hot water turned off until we can shed some light on this. Anyone have any ideaa what this could be and is it ok to leave the hot water on or not?

How would I test that? Would a thermostat failure make a noise like that?
