Standing water in toilet waste pipe

I had a peek into my 110m toilet waste pipe under the floor boards from a boss entry. There is standing water inside (maybe about 20% capacity of the pipe). Is this normal or does it suggest incorrect gradient or blockage?

Not normal, and yes, would indicate a partial blockage or backfall. Suggest you investigate further, may be worth checking external runs are clear before doing anything else.

The gradient of the pipe is pretty much bang on level. Would this be a problem?

Yes. Needs ideally a 1:40 fall, 1:100 at an absolute minimum.
