I am installing shower today and I put a piece of ply behind the plasterboard for more support.

So, i have tile, adhesive, plasterboard, plywood in that order

How do I fix the shower rail? Do I use a plug and screw through the whole lot or just drill a pilot hole in tile to width of the screw, and then screw through into plywood (with no plug)

Cheers

Just drill through the tile and screw straight into the plywood. Make hole slightly larger than screw diameter, use stainless steel screws and I always squeeze a small amount of clear silicone into hole before inserting screw.
