Help on valve settings

First things first, get the PTFE tape off the threads of the unions of the 3 way valve. It can actually make the joints leak rather than seal it up, one on the right is weeping by the looks of it. Tape should only be used on the olives, if they are being re-used.

Without knowing what the gate valves feed (red handles) then it's nigh on impossible to tell you which one should be open/closed/part open. The only ones to be certain about are the ones above and below the pump which should be open and the bypass which should be partly open but that needs to be set up properly.
 
Unless you have trvs on every radiator you don't need the bypass, the gate valve on the heating should not be there so leave that fully open.
 
If it is possible take a panoramic picture, because it is not clear that in the left side.

Then close the one on the left hand side of the pump and open the one on the right.
 
With reference to the red gate valve heads in your most recent photo, from left to right:

Fully open
Fully closed
Fully open

And set the timer so that the hot water comes on immediately before or immediately after the central heating comes on (logic being that water will take the easier route, and going around the cylinder coil is much easier than going around the radiator circuit).

Edit: When I say fully open, turn them all the way open and then back off 1/8 of a turn to help prevent them sticking.
