We are looking at possibly converting our garage (well, dumping ground) into a useable living / working area. There are 2 man-hole covers within the existing garage area... will we be able to block these off or are we scuppered?

It’s very unlikely you’ll be able to just block them off; do you know if they are foul or surface water drains? Do they serve just your property or your neighbors as well? You can get double sealed covers which will allow you to have them inside your conversion but you must maintain access to clear blockages. But, TBH, do you really want two inspection hatches in your conversion! I would budget to re-route the drain but you need to asses what’s possible by looking at the drainage route & who/what it serves.

Converting a garage is also notifyable building work, see here for more detail;
