Can I use MDPE pipe for mains water within the house (under floors, behind covers) up into the loft, to feed an unvented cylinder?

Or do I have to use PEX if I do not want to use copper (because of cost)?

No reason why you shouldn't, except that it's relatively bulky, especially when it comes to fittings. By the time you've purchased fittings and inserts, the cost of PEX and copper is fairly similar.

Its probably cheaper than copper or PEX and the advantage is that it will probably be larger bore.

If I was doing it for myself I would run it straight in and up all in one bit for the maximum flow rate.
