Made a square roof vent in my ensuite. Do i have to line the inside of the wooden upstand with plasterboard or can i plaster straight onto it?

do not plaster onto wood.

wood expands and contracts, plaster don't.

I plastered onto plywood in the bathroom 20 years ago, Nothing has happened.

You may get away with it if you paint a coat or two of waterproof PVA on the wood before plastering? The first coat seals the wood a bit and the second helps the plaster grip.
