Do you have zonal heating , a thermostat for each upper floor ?

Might be zones

Might be balancing

You can tell us more about your controls, and valves, and show pics.

This is the thermostat on middle floor with no heating, I have exactly the same on the top floor on exactly the same settings and they are on

They will be fitted on your central heating Pipework . squarish shaped Box ( metal or white plastic ) with electric cable coming out of them ,to loosely describe them. You will have a number of them !

It is indeed. How many of them can you find ? One of them will operate the rads that do not heat up and the pipe going into it will be hot and the pipe going out will be cold further along its length.

There only one downstairs next to the ufh manifold and its hot both sides, I'm certain this is the only one in the house

It can't be ,look elsewhere .Do you have a hot water cylinder ? If so look there.

This is the only cupboard in the house, I have a combi and no hot cylinder. The house is only 5 years old and every available space is a room such as the top and middle floors, there are no other voids or cupboards for another valve

No two way valves under the combi?

You would think the stats on the middle and top floors would drive two way valves?
