We have a karndean click flooring in our dining room that has been down for about 6 years, it's laid over 6mm ply on top of floorboards.

One of the planks in the centre of the room has developed a dip and this has led to the edge coming away slightly and the top layer chipping off.

I've got a few spare planks in the garage. Is there a way to remove and replace the plank without having to pull the whole floor back to the wall?

You may get away with cutting the damaged plank out, trimming the click tongue off the replacement plank and glueing it in place direct to the floor, but lifting planks back to the replacement area is the correct way to tackle this.
