Screw Holes for metal/steel.

Was putting together a shoe rack and seat, the bottom being of a metal/steel material, I noticed that putting together the bottom on opposite ends of the screw holes one was directly round and the other being round too but with a bit cut out at the side, my question is: does it matter which end the screw goes into first as the piece could be screwed in by turning it both ways.

The ends that have the cut out must be there for a reason.

Any help appreciated.

Does the bolt have s small knib under the head , slot could be there to locate knib and stop bolt rotating.?
