What distance is required between joists and subfloor?

Should sub floor be treateed with anything? It is currently bare clay.

150mm minimum from u/side joists to solumn. Solumn - 50mm concrete, dpm, 50mm sand. This is my preferred solumn make-up. hope this helps

Just to clarify, its sand on clay, dpm on sand , concrete on dpm. ?
