Can someone please tell me what this isolator valve does?

Always wondered and never truly found out what this isolator valve does in the airing room next to the h/w tank.

Should it be open or closed?

Unless you have Thermostatic rad valves on ALL rads.
Then it should be opened approx 3/4 turn.

Got no thermostatic rads at all in the house so does that mean it should be closed?

I'd leave it be if I were you, all the rust marks and crusty bits the chances are it will leak if you touch it.

As you have no TRV's then it can be closed as suggested.
