Do you mean the return from the bypass? If so this is normally connected to the main return after the cylinder return, i.e nearer the boiler.
The return from the cylinder should be the last one before the boiler otherwise you could end up with reverse circulation. But as already stated it depends on whether you mean an actual bypass or not.
