Radiator balancing, when to check temp

Hiya all.

So, I'm after balancing my rad and have my thermometers ready. after reading through some posts it seem relatively straight forward but I'm unclear if you need to check the temps while the heating is warming up from cold or not.

The reason I ask is that while checking the temp on the first rad (nearest the boiler) there's was a 20oC diff but after choking the next couple which were around the same diff I went back to the first and the temp diff was 8oC.

Can anyone tell me if I need to check the system while its heating rather than after its been on for a while please?

Not sure if makes a diff but its a vokera compact 24 sealed combi

Oh, probably oh worth mentioning that my boiler is downstairs and the locks idle valves take 4 & 3/4 turns to go from fully close to full open
