replacing ceramic floors

My house has a 20 year old ceramic tile floor (approx 400 sq.ft.) that I would like to replace. There are two layers of 1/2" plywood under the concrete setting material. Can I use concrete board ("WonderBoard") directly on top of the old setting material? Or should I take up the two layers of plywood, and rebuild from the joists up? Thanks
