I’m wanting to replace vented dryer with heat pump so vent will be obsolete. Is it ok to put a piece of ply over the internal hole and just leave the grill over the external side? Think that’s as much as we could do as DIY

Stuff it with glass-wool insulation from the inside, and leave the grill on. (y)

Rather than ply, you could screw a plastic hit-and-miss vent over the inside. It will not be damaged if it gets damp, unlike ply, and you can easily open it if you ever want extra ventilation, for a hot appliance like an oven or freezer, or for an aircon unit, or if vented driers ever come back into fashion (they are remarkably simple and cheap)

On the outside, fit a louvred grill that will throw off rain, not one with flaps that rattle

Preferably one with an insect screen, unless you don't mind your wall cavity becoming a nature reserve. :giggle:
Won't the vent be lined? Mine is so all I did was stuff a load of loft insulation into the vent and sealed it on the inside.

Some folk just like to overthink stuff (me included).

I'd stitch out the affected bricks and re-do the masonry. Glass wool stuffed up to the vent pipe is a simple DIY remedy.
