I have a 3 port ufh manifold, taking temp of all flow and returns, the 1st on manifold has a flow 4 degrees lower than other although return is similar, tried playing with valves but still cant get flow up to near others, any thoughts?

Blockage or air in the loop. You could try flushing the loops through and see if it improves. Don't use the boiler fill. You'll need to connect a hose a per instructions for your manifold.
