Using interior filler on MDF

Neither, use dec caulk it's flexible, quicker and easier to apply.
Or use car filler, dries quick and sand down well.

I always use 2part polyester filler. The sort thats used for bodywork
As said, it dries very quick, fairly crack resisitant, and sand down easily.
