filling ahole in the tile

Is it that the hole is no longer holding the fixing secure for the rail or just a matter of making it look neater?
If the latter, grout it with same colour as tile.
If it's a loose fixing, it is likely you will need to re-drill for larger sized plug an re-fit rail.
