Fitting combi on bbu system, question

Gravity hot water pumped central heating, combi upstairs: after taking out heat exchanger, can i connect up flow and return on 22 central heating system, or will this cause a negative spot? and can I then break in upstairs on the flow and return to the combi boiler, or do I have to take the 22 flow and return in the back boiler void up to the upstairs combi?
