How do I clear my airlock?

I've just fitted a new Honeywell mid-position valve to my heating system.
The new valve came already attached to the three-way pipe fittings so I replaced the whole apparatus - which meant I had to drain my hot water cylinder.
In doing this, the expansion tank emptied so I bled every radiator before refilling the system.
I restarted the boiler by setting the HW to ON with the boiler on Max, and CH off. I switched the boiler on and it fired up - then after a couple of minutes it cut out.
The Grundfos Selectric pump is now making a strange noise, like a steam train on idle, and it gets hot but the pipes either side of it remain cold.
Consequently I don't have any hot water or heating, and I'm worried that the pump is running dry so I've switched off the boiler).
I opened up the bleed screw on the pump and the pump is definitely turning but it seems no water is being fed into it to actually pump.
What is the cause of this and how do I fix it? I'm thinking the cause might be air trapped in the system - most likely between the header tank and the pump?

How exactly do I bleed the pump? Through the silver screw? I thought I had bled it but maybe I haven't...
