I want to replace my immersion heater and am asking what is the best sealing method to use.?

how does that affect undoing it when it needs replacing in X years time and the thread is seized?

I was toying with the idea of PTFE tape on the threads and flange, not as a sealant, but an anti-seize.

Assuming that the flange and thread on the cylinder are in good condition, put it in dry with the new fibre washer. No sealant, you don't need it. Then it can come out easily enough.
