Screeding a beam and block floor

We recently had to get rid of our builder because of a few issues and dramas and are now looking to lay the floor of our extension ourselves. It’s needs to be done ASAP as the kitchen fitters are coming in a few weeks.

He left a pile of insulation we’ll be putting down but I wanted to ask if all the gaps need filling first or should we just put the insulation straight down over them? Once the insulation is down, does there need to be a polythene lining or just screed straight on top?

Any advice welcome!

Insulation straight on top, a thin vapour barrier on top to prevent the screed bleeding into the gaps between the insulation boards.
