If swapping a pair of taps on a basin with flexis with iso valves in the supply. 15mm with iso to 1/2 female. Would you usually just isolate and undo the 1/2 female end fit taps and tighten back up?

You need to be careful if the female end of the flexi is straight onto a compression end of the ISO valve, if the ISO isn't the type that has a 1/2" BSP flat end on it.
If it's the former then they're designed to be used with olives and compression nuts and the sharp edge can easily cut into the rubber seal in the female end if too tight. The only issue with changing it now to say a tap tail adapter is that there won't be a compression nut or olive any more. You can buy another compression fitting and just use the nuts and olives from that, should be the same thread and then use a pair of these for the flexi's to fit onto.
There is a way I use the ISO's without the above and that's by using a flat file on the end of the brass ISO, to widen the mating edge, so it doesn't cut into the rubber seal but I have the tools to do all of that.

Ahah, ok, yup just shut off the valve, loosen the tap end, swap the taps and then reconnect, hand tight then 1/4 turn. Switch back on and check for leaks.

No need to swap the washers in the flexis or anything?
Shouldn't need to , give them a check and as long as they aren't perished or cut then they should be fine. You can turn them around if they are overly marked.
If needed could you put new washers in from another Flexi?
yes, as long as they are the same size.
