I'd have thought those who want a second referendum would welcome the EU election as a barometer of public mood on Brexit. If the exit parties do well, it suggests people want to leave, if the remain parties do well it suggest remain. If the BRINO parties do well its suggest we want a compromise.

Could you define a 'Brexit party'?
Obviously the so named party is one (along with UKIP), and ChangeUK, the Greens, SNP and the Lib Dems are remain...
But what would you say about the usual suspects (Cons and Labour)?
And what percentage of the vote would you consider to be a 'barometer'?
Lets pluck a figure out of the air - 48% will do...
If you can define the Brexit parties and they got this figure or lower, would it be a signpost for another referendum?
And of course what about the turnout? Does the percentage count? Or the actual numbers voting? Or the number of MP's elected?
(But aren't parliamentary elections supposed to be about a range of issues?)
All this of course has the big caveat that EU elections, like local elections and the 2016 referendum, are often used as an opportunity for a protest vote...
But it must be noted that at least the EU elections (being a form of PR) are more democratic than our antiquated FPTP system!
(if we had a UK parliament elected by PR we probably wouldn't be in our current predicament!)

You are, as Ellal suggests, disregarding other issues on the manifestos of the various parties. Although we all recognise that not all parties have manifestos.
