i am changing my water mains pipe, and its position. It was in the back of the garage however I am changing it to the front of the garage. Now it has to go over a door in garage. Will this have a effect on pressure in anyway?

Yes, because every metre of pipework and bend or change of direction adds extra resistance and therefore pressure drop, but not enough to worry about in the context of your question.

No, in the context of your fears.

Sending water upwards loses pressure but that is reinstated by gravity when it comes down again.

As long as it goes up and then comes down again.

Properly pipe sized the added flow resistance will be negligible.
