Glow worm betacom 28a. I have done as much googling and YouTubing as I can guys but still no joy

So if you turn that blue valve then no water flows in to pressurise the boiler? Oh and that valve is on the wrong end
 
OK so you need to investigate that valve/filling loop 1st to see why it's not letting water by and filling the boiler. Take one of the ends off and see if the valve's working.

No water is coming out when valve open, to I need to screw the screw in picture?

If you open the hot water taps are you getting water out, albeit cold?

If so, the blue handle on the valve may be worn and isn't turning the internal ball to open the valve. You need to loosen the wee screw inside the blue handle, pull it off and see if you can turn the brass square underneath it to open the valve, with an adjustable spanner or pliers. Careful as you could get wet very easily.

Don't touch the valve in your 2nd pic

Just for your info ... strictly speaking it's the wrong valve (that's an appliance valve) and the check valve on the other end of the flexi should be where the blue valve is and vice versa.
